AFB is the most devastating bacterial disease affecting honeybee brood worldwide caused by the spore-forming, Gram-positive bacterium Paenibacillus larvae. In areas where disease incidence is high, oxytetracycline is currently used to prevent and control AFB in honeybee colonies as an alternative to the burning of infected beehives. However, tetracycline-resistance (TcR) bacterial strains have been detected in USA, Canada and Argentina. The Epsilometer test - ETest® - is a quantitative technique for determining Minimum inhibitory concentrations (MICs) of antibiotics and is a rapid alternative to conventional agar or broth dilution MIC determinations. ETest is calibrated to give performance equivalent to US Clinical Laboratory Standard Institute (CLSI) reference methods. The aim of this work was to evaluate the accuracy of the ETest for determining the quantitative susceptibilities of P. larvae to tetracycline in comparison with the agar dilution method. A collection of 17 P. larvae strains from different geographical areas with different tetracycline susceptibility was tested by using two different culture media, MYPGP agar and Oxoid Iso-Sensitest agar® to perform comparisons between ETest and agar diffusion method for MIC determinations. P. larvae strains ATCC9545, NRRL B-3555, CCM38 and ATCC14154 and Staphylococcus aureus ATCC29213 and ATCC25923 were also included as references and quality controls, respectively. Results showed that P. larvae growth was enhanced by using Iso-Sensitest® agar and no differences were found when compared ETest with agar dilution in this medium, but when using MYPGP medium discrepancies were found between MIC values obtained with ETest compared with agar diffusion, particularly in resistant strains. In conclusion, the ETest® is a reliable and rapid method for testing P. larvae susceptibility to tetracycline only if it is used in combination with the Iso-Sensitest® agar. INTRODUCCIÓN La loque americana ocasionada por Paenibacillus larvae es la enfermedad más grave de origen bacteriano de la etapa larval de las abejas. Está difundida a nivel mundial y, en algunos países, se controla mediante aplicaciones del antibiótico oxitetraciclina. El Etest® (Epsilometer-test) es una técnica cuantitativa para la determinación de la Concentración Inhibitoria Mínima (CIM) de antibimicrobianos como una alternativa rápida a las determinaciones convencionales de CIM mediante el empleo de diluciones en agar o en caldo de cultivo. OBJETIVO Evaluar el desempeño diagnóstico del Etest comparándolo con el método de dilución en agar para la determinación cuantitativa de la susceptibilidad / resistencia de Paenibacillus larvae hacia tetraciclina y correlacionar las CIM obtenidas por ambos métodos empleando dos medios de cultivo : MYPGP e ISO Sensi-Test agar. MATERIALES Y MÉTODOS Método de dilución en agar Oxoid Iso-Sensitest agar® Determinación de las CIM Antibiogramas mediante prueba de difusión en disco Agar MYPGP Etest® (Epsilometer -test) RESULTADOS No se encontraron discrepancias entre los valores de CIM obtenidos por dilución en agar y los obtenidos con ETest en el caso del agar Iso-Sensitest® (Gráfico A) pero cuando se empleó MYPGP se encontraron diferencias entre los valores de CIM obtenidos por Etest comparados con los obtenidos por dilución en agar, particularmente en el caso de las cepas resistentes (4 errores minor en cepas que pasarían de resistentes a intermedias) (Gráfico B).
