SEÑAL EGFR Expresión en casi el 100% canceres cabeza y cuello Slide 3 EGFR signaling The EGFR is activated by ligands binding to the extracellular domain.1,2 Following binding, the receptor monomer forms a dimer with another HER receptor to form either a homodimer (with another EGFR receptor) or a heterodimer (with another receptor of the HER family).2 Dimerization is followed by cross phosphorylation of the tyrosine residues on the receptor pair, which then form docking sites for transient signaling complexes.2 Dissociation of these complexes results in the stimulation of signal cascades, such as the mitogen-activated protein kinase (MAPK) pathway and the anti-apoptotic kinase Akt, leading to gene activation and subsequent cell responses.2 The EGFR signal is inactivated by internalization and then either degradation or recycling of the receptor.2 REFERENCES: Baselga J. RT was administered according to one of three fractionation regimens, according to investigator preference: once-daily 70 Gy, 35 fractions; twice daily 72–76.8 Gy, 60–64 fractions; or concomitant boost 72 Gy, 42 fractions. Patients were stratified by: KPS 60%–80% vs 90%–100%; N0 vs N+; T1–3 vs T4 and RT fractionation regimen. ERBITUX was administered as a 400 mg/m² initial dose 1 week prior to RT followed by subsequent doses of 250 mg/m² per week. Patients were followed for a total of 5 years, by physical examination and radiographic imaging (every 4 months for the first 2 years and then every 6 months for the next 3 years).
